Raw Material Selection & Grinding

Production uses high-quality dried broccoli seeds or fresh broccoli heads as the main raw materials. First, the raw materials are selected, cleaned, and pre-treated to remove impurities and ensure the quality meets production requirements. The treated broccoli raw materials are then crushed using a pulverizing device to achieve a particle size of 30-60 mesh. This effectively disrupts the plant cell structure, allowing for the full release of glucoraphane and related enzymes, thus improving the efficiency of subsequent enzymatic hydrolysis.

 

Enzymatic Hydrolysis

The crushed broccoli raw materials are mixed with 1.5-2 times their weight of pure water, and the pH of the system is adjusted to 4.1-4.5 using a 5% hydrochloric acid solution to establish an acidic environment suitable for the myrosinase reaction. Then, 1g of crude myrosinase (crude black myrosinase) is added per kilogram of raw materials, and the enzymatic hydrolysis reaction is carried out at 31-34℃ for 1-3 hours. Glucosamine undergoes hydrolysis under the action of myrosinase, transforming into active sulforaphane.

 

Freeze Drying and Material Stabilization

The broccoli solid-liquid mixture after enzymatic hydrolysis is freeze-dried to remove moisture at low temperatures, ensuring the final powder moisture content is below 5%. Freeze-drying effectively reduces the degradation of heat-sensitive active substances while improving material stability, providing suitable conditions for subsequent organic solvent extraction. The freeze-dried powder then proceeds to the next extraction step.

 

Solvent Extraction and Crude Product Preparation

The freeze-dried broccoli powder is extracted with pure acetone. Pure acetone is added three times by weight in three separate extractions at room temperature, with a total extraction time of 4–8 hours. After extraction, plant residues are separated by filtration, and the three acetone extracts are combined and concentrated under vacuum at 35–40°C to remove the solvent, yielding crude sulforaphane.

 

Multistage Column Chromatographic Purification

The obtained crude sulforaphane was further purified using silica gel column chromatography. During purification, a mixture of ethyl acetate and ethanol was used as the eluent, and the target components were separated by gradient elution. The fraction containing sulforaphane was collected and concentrated under vacuum at 35–40°C. Subsequently, the concentrated sulforaphane was dissolved in methanol, filtered through a 0.2 μm microporous membrane, and further refined using a 20–40 μm C18 silica gel column. Gradient elution was performed using methanol-water systems of different concentrations to remove impurities.

 

Vacuum Drying & Final Product

The sulforaphane fraction purified by the C18 column was collected and subjected to desolvation under vacuum drying at 35–40°C to finally obtain high-content sulforaphane powder. After quality testing, different specifications of the product can be prepared according to customer requirements, including standard specifications with sulforaphane content of 1%, 5%, 10%, and 13%, or customized according to customer requirements.
