Raw Material Acceptance and Pretreatment

Before production starts, the purchased dried Eucommia ulmoides leaves undergo quality inspection to ensure their chlorogenic acid content is within the range of 1.0% to 5.0%. The qualified leaves are then pulverized to a suitable particle size and precisely weighed according to the feeding plan to provide a uniform raw material base for subsequent extraction.

 

Dynamic Water Extraction

The weighed Eucommia ulmoides leaves are placed in the extraction tank, and purified water is added at a ratio of 10 to 14 times the weight of the medicinal material. The pH of the extraction system is adjusted to a slightly acidic range of 6 to 7 using acid. Stirring or dynamic circulation is started, and the extraction temperature is stably controlled between 60°C and 80°C. Each extraction lasts 0.5 to 1.5 hours, and 1 to 3 extractions are repeated based on the target transfer rate. After each extraction, the extract is filtered and collected, and the extracts from all extractions are combined and transferred to the next process.

 

Chitosan Clarification and Coarse Filtration

The combined extracts were transferred hot into a settling tank, and chitosan, a natural clarifying agent, was added at a precise amount of 0.4% to 0.8% of the drug mass. The mixture was allowed to settle for 1.0 to 2.0 hours at a temperature of 50°C to 70°C. Chitosan effectively removed large molecular impurities such as proteins, pectin, and some pigments from the extract through flocculation, significantly reducing the viscosity of the solution. After settling, the pretreated solution was coarsely filtered through a membrane or filter cartridge with a pore size of 50 to 100 micrometers to prevent clogging of the subsequent resin column.

 

Macroporous Adsorption Resin Chromatography Purification

Non-polar or weakly polar macroporous adsorption resins were selected, and the column diameter-to-height ratio was ensured to be 1:4 to 1:8 during packing. During the sample loading stage, the feed flow rate is controlled at 3.0 to 5.0 column volumes per hour, and the adsorption loading is dynamically adjusted according to the ratio of medicinal material mass to column volume of 1:1 to 1:4. After sufficient adsorption, the resin column is first rinsed with 1.0 to 2.0 column volumes of purified water at a flow rate of 3.0 to 5.0 column volumes per hour to remove highly polar unadsorbed impurities such as sugars and inorganic salts. A 10% to 30% aqueous ethanol solution is used as the eluent at a flow rate of 4.0 to 6.0 column volumes per hour for desorption and elution. The eluent rich in chlorogenic acid is collected, and the initial and final impurities are discarded.

 

Voltage Concentration

The collected ethanol eluent is transferred to a vacuum concentration device. The concentration temperature is controlled between 50°C and 70°C, and the vacuum degree is maintained at a high vacuum condition of -0.07 MPa to -0.09 MPa. The ethanol in the eluent is recovered and evaporated, and the concentration continues until a clear paste with a relative density of 1.03 to 1.05 is obtained. Low-temperature vacuum operation effectively protects the heat-sensitive chlorogenic acid from high-temperature damage, while simultaneously recovering the ethanol solvent for recycling.

 

Spray Drying and Finished Product Packaging

The concentrated paste is pumped into a spray drying tower. The inlet temperature is set to 150°C to 170°C, and the outlet temperature to 60°C to 80°C. The paste is dispersed into fine droplets by an atomizer and comes into instantaneous contact with the hot air, causing rapid evaporation of moisture and obtaining a dry crude chlorogenic acid powder. After cooling, sieving, and mixing, the dried powder is packaged according to specifications (both inner and outer packaging). After passing quality inspection, it is stored as the finished chlorogenic acid powder.
