Mangiferin is a naturally occurring C-glucosyl xanthone, mainly derived from the leaves, skin, and core of mango (Mangifera indica), with the highest content in mango leaves reaching 6.78% (w/w). However, Mangiferin faces two major technical challenges in practical application: extremely low water solubility (0.111 mg/mL) and poor oral bioavailability (only 1.2%). These defects are mainly attributed to its molecular structure characteristics: the molecular weight is 422.34 g/mol, the log P is -0.65, it lacks sufficient lipophilicity to penetrate the biological membrane, and it is difficult to dissolve in the aqueous environment due to the presence of C-glucosidic bonds. Therefore, targeted formulation strategies need to be developed in different application areas to overcome these limitations.
