Raw Material Alkalization And Mixed Solvent Hot Reflux Extraction
The dried roots of Aconitum carmichaelii were pulverized and placed in a hot reflux extraction tank. Sodium hydroxide was added for alkalization to fully release the alkaloids. Then, a mixed solvent of methanol and ethanol (volume ratio 2:1) was added, and dynamic hot reflux extraction was performed at 40℃ to 50℃ for 5 hours. After extraction, the mixture was filtered, and the extract was concentrated under reduced pressure to recover the solvent, yielding a crude extract of Aconitum carmichaelii.
Acidification of crude extract and precipitation of free alkali
The crude extract was adjusted to acidity with acid, causing the alkaloids to transfer to the aqueous phase as salts. Simultaneously, insoluble impurities were removed by filtration, yielding an acidified solution. The acidified solution was then adjusted to alkalinity with alkali, causing the free alkali of Aconitum carmichaelii to precipitate. The precipitate was collected by solid-liquid separation, achieving preliminary separation of the target component from impurities.
Crystallization and recrystallization purification
