This provides abundant substrates for subsequent enzymatic hydrolysis, improving extraction efficiency and product yield.

First Stage Complex Enzymatic Hydrolysis

After autolysis, the yeast milk is cooled to 60–70°C, and the pH is adjusted to 6.5–7.0. Yeast extractant, papain, and alkaline protease are then added, and hydrolysis is carried out at a constant temperature for 8–12 hours. This stage primarily utilizes the synergistic effect of multiple proteases to gradually break down large protein molecules in the yeast into smaller peptides and free amino acids, improving protein utilization and promoting the release of umami precursors.

Second Stage Enzymatic Hydrolysis

After the first stage of enzymatic hydrolysis, the resulting hydrolysate is cooled to 40–50°C, and the pH is adjusted to 4.5–5.5. Then, acidic protease, neutral protease, and nuclease are added, and the enzymatic hydrolysis reaction continues for 10–20 hours. This stage further degrades residual proteins, peptides, and nucleic acid components. Nuclease promotes the hydrolysis of nucleic acids to generate flavor nucleotides, enhancing the umami flavor of the product and improving its solubility and nutritional composition.

Flavor-Directed Enzymatic Hydrolysis Optimization

After the second stage of enzymatic hydrolysis, the hydrolysate is reheated to 50–60°C, and the pH is adjusted to 5.5–6.5. Glutaminase, flavor protease, and deaminase are added, and the targeted enzymatic hydrolysis reaction continues for 10–20 hours. This stage mainly promotes the further conversion of flavor precursors, increases the content of umami amino acids such as glutamic acid, optimizes peptide composition, and enhances the natural umami flavor and overall flavor quality of the yeast extract.

Enzyme Inactivation and Solid-Liquid Separation

After flavor enzymatic hydrolysis, the hydrolysate is heated to 75–85°C and maintained for 0.5–2 hours for enzyme inactivation treatment, completely deactivating all enzyme preparations, terminating the enzymatic hydrolysis reaction, and ensuring stable product quality. After inactivation, the material undergoes solid-liquid separation to remove undissolved particles and cell residues, obtaining a clear and homogeneous yeast extract, providing a stable raw material for subsequent concentration and drying processes.

Concentration and Spray Drying

The separated yeast extract is concentrated according to process requirements to increase the soluble solids content. Then, a spray drying process is used to rapidly dry the concentrate into powder.
