1.What is Sesamin ?
Sesamin Extract exists in the root of Araliaceae plant Sessiliflorus [Acanthopanax sessiliflorus (Rupr. et Maxim.) Seem.], in the seeds or seed oil of Sesamum indicum DC. Thunb.) Steud. Xybe and other plants. Its properties are white needle-like crystals, easily soluble in chloroform, benzene, acetic acid, acetone, slightly soluble in ether and petroleum ether. Insoluble in water, alkaline solution and hydrochloric acid.
Sesamin is a natural component contained in sesame seeds. It is the lignan with the highest content in sesame seeds. Sesamin contains about 0.3-0.5% sesamin. It has various health benefits, including cholesterol-lowering activity, hypotensive effect, anticancer potential, anti-aging effect, immunomodulatory activity, hypoglycemic effect, antithrombotic effect, hepatoprotective effect, and antioxidant effect. 5.Assay
A method for determining the sesamin content in sesame seeds, comprising the following steps: (1) preparing a sesamin standard solution: weighing 10 mg of a sesamin standard product, adding anhydrous methanol to dissolve and diluting to 50 mL, and then using a 0.45 μm filter membrane Filter to obtain the sesamin standard solution of 0.2mg/mL;
(2) Draw the standard curve of sesamin: take 20, 10, 5, 4, 2, 0.8mL of the sesamin standard solution obtained in step (1), add 0, 10, 15, 16, 18, 19.2mL of anhydrous methanol respectively , Shake well to prepare sesamin standard methanol solutions with gradient concentrations of 200, 100, 50, 40, 20, 8ug/mL respectively, use a liquid chromatograph to carry out chromatographic analysis on the above solutions, and draw the sesamin standard according to the measurement results curve (see Figure 1 for the liquid chromatogram of the sesamin standard solution), and obtain the linear relationship equation between the sesamin content and the peak area: A=587.525675x-4.531582, where A is the peak area, and x is the sesamin content;
(3) Prepare the sample solution to be tested: use Yuzhi No. 11 sesame seeds as the sample to be tested, crush the sesame seeds to be tested with a universal grinder for 30 seconds, and then sieve them through a 0.35 μm sieve to take 0.2 g of the obtained sesame powder sample , adding 5 mL of ethanol with a concentration of 100%;
(4) Ultrasonic treatment and filtration: ultrasonically treat the mixed solution obtained in step (3) at 30-60°C for 20-50 minutes, and then filter the ultrasonically treated solution with a 0.45 μm filter membrane; ultrasonic treatment The conditions are: use a digitally controlled ultrasonic cleaner with an ultrasonic frequency of 40KHZ and a model number of KQ3200DE, and perform ultrasonic treatment at an ultrasonic power of 70-100W.
(5) Detection: Take 9 μl of the filtrate obtained in step (4), use a liquid chromatograph to inject a sample of the filtrate for detection, and calculate the content of sesamin in the sesame seeds to be tested according to the sesamin standard curve and the obtained linear relationship equation .
