How to make Sophora Alopecuroides Extract Cytisine Powder
Supercritical CO₂ Extraction
Select qualified Pterocarya stenoptera seeds, remove impurities, and pulverize to 40-60 mesh. Weigh and add to a supercritical extraction vessel. Start the refrigeration and heating devices, turn on the compressor pump to pressurize to 21-22 MPa, and pump in liquid CO₂ for circulating extraction. Under supercritical conditions, CO₂ carries cytisine into the separation vessel. After depressurization and desorption, collect from the bottom of the separation vessel to obtain crude cytisine.
Macroporous Resin Separation and Purification
Place the crude product in a mixing tank, add 10-20 times the amount of acidic aqueous solution to adjust the pH to 1-3, stir to dissolve, and filter to remove insoluble matter. Add alkali to the filtrate to adjust the pH to 7.5-8.5 to release cytisine, and then pass it through a pretreated macroporous adsorption resin column for adsorption. After adsorption, wash with water until neutral, then elute with 4-7 times the amount of 60%-90% ethanol. Collect the eluent and transfer it to a vacuum concentration tank. Recover the ethanol under vacuum and concentrate to a thick paste to obtain a fluid extract.
Crystallization and Refining
Acetone is added to the fluid extract, and the mixture is heated under reflux to dissolve completely. The mixture is then slowly cooled to precipitate crystals. After filtering out the initial crystals, the dissolution and crystallization steps are repeated 2-3 times. The resulting white crystals are filtered out, dried at 40-50°C to remove residual solvent, pulverized, sieved, and thoroughly mixed before being packaged according to GMP requirements to obtain the finished Sophora Alopecuroides Extract Cytisine Powder product.
