The Polygonum cuspidatum powder is added to 10 times its volume of purified water and treated under 300 watts of ultrasound for 10 to 15 minutes to initially break down the cell walls using ultrasonic cavitation. Subsequently, 1% to 5% (by weight) of cellulase is added, and the pH of the reaction system is precisely controlled between 4.5 and 5.5 using an acidity adjuster. The enzymatic hydrolysis reaction is carried out at a constant temperature of 40 to 60°C for 60 to 120 minutes. After enzymatic hydrolysis, the precipitate is collected through solid-liquid separation.
The precipitate obtained after enzymatic hydrolysis is transferred to a supercritical extraction vessel. The extraction vessel pressure is set to 20–35 MPa and the temperature to 40–55 °C; the desorption vessel pressure is set to 5–7 MPa and the temperature to 40–55 °C. The circulation system is started, and a mixture of anhydrous ethanol and 2-propanol at a volume ratio of 7:3 to 8:2 is pumped in as a modifier. After continuous extraction for 60–120 minutes, the desorption vessel pressure is adjusted to 5–6.5 MPa and the temperature to 40–50 °C, and the extract rich in resveratrol is collected.
The obtained extract is concentrated under reduced pressure at 40–50 °C and a vacuum of -40 cmHg to -60 cmHg. The anhydrous ethanol and 2-propanol mixture modifier is recovered, yielding a concentrated filtrate containing resveratrol. The concentrated filtrate was loaded onto an activated and cleaned macroporous adsorption resin column for chromatographic separation. Utilizing the resin's selective adsorption capacity for components of different polarities, resveratrol was adsorbed onto the resin column, while impurities flowed out with the eluent. An appropriate concentration of eluent was used to desorb the resveratrol from the resin, and the high-purity target fraction was collected.
The high-purity eluent after chromatographic separation was collected and concentrated again under reduced pressure to remove the solvent, yielding a concentrated solution. This concentrated solution was transferred to a freeze-drying apparatus, where water sublimated at low temperature, ultimately obtaining a dried Resveratrol powder.
