Raw Material Pretreatment

Take rhubarb raw material and pulverize it to 20-80 mesh using a pulverizer. In conventional production, the pulverization size is controlled at around 40 mesh to increase the specific surface area of ​​the material and improve the efficiency of subsequent enzymatic hydrolysis and extraction.

Enzymatic Hydrolysis

Add 1-2 times the weight of water (preferably 1.2 times) to the pulverized rhubarb raw material for wetting. Then add 0.1%-0.5% of a compound enzyme (preferably 0.3% glucosidase) and hydrolyze at 30-50℃ for 2-5 hours (preferably 3 hours at 40℃). The enzyme used can be one or a combination of cellulase, glucosidase, rhamnosidase, and pectinase.

Organic Solvent Extraction

After enzymatic hydrolysis, add 3-10 times the weight of organic solvent (preferably 6 times) to the hydrolysate. The organic solvent should be a 70%-90% methanol aqueous solution or an ethanol aqueous solution (preferably 80% ethanol aqueous solution). Extraction is carried out at 70-80℃. The extraction is performed 1-3 times (preferably 3 times), and the extracts from each extraction are combined. After extraction, concentrate the extract to a specific gravity of 1.05–1.20, preferably to 1.15–1.20 (e.g., 1.17).

Alkali-Alcohol Dissolution

Add 20–50 times its mass of an 80%–90% methanol or ethanol aqueous solution (preferably 40 times its mass of an 85% ethanol aqueous solution) to the extract, and simultaneously add 20%–30% of the extract's mass of an alkaline solution (preferably a 25% sodium hydroxide solution). Dissolve at 75–85°C for 2–5 hours (preferably 3 hours at 75°C). The alkaline solution can be one or a combination of sodium hydroxide, potassium hydroxide, sodium carbonate, potassium carbonate, and sodium bicarbonate. After dissolution, filter. Repeat filtration on the residue and test the aloe-emodin content until the aloe-emodin content in the residue is ≤2.0%. Collect and combine the filtrates.

Acid Precipitation and Purification

The combined filtrates are allowed to cool to room temperature, and acid is added to adjust the pH to 3-5 (preferably pH 3). Precipitation is allowed to occur for 0.5-2 hours (preferably 0.5 hours). One or more combinations of sulfuric acid, hydrochloric acid, citric acid, glacial acetic acid, and oxalic acid can be used. After precipitation, the precipitate is filtered and washed with water, alcohol, dichloromethane, or n-hexane. Finally, it is dried to obtain the Aloe Emodin Powder product.
