Raw Material Pretreatment
Take qualified dried rhizomes of rhubarb (Rheum palmatum or Polygonum cuspidatum) and place them in an industrial-grade drying oven at 110–130℃ for drying. The dried rhizomes are then pulverized to a fine powder of 100–120 mesh. The pulverized powder is then placed in a multi-functional extraction tank and pretreated using steam distillation. The distillation temperature is controlled at 50–80℃, and distillation is continued for 2–3 hours.
Extraction and Primary Separation
Take 30–40 parts by weight of the refined powder and add it to 150–200 parts of an ethanol aqueous solution (50%–70% ethanol). Extract by reflux at 60–80℃ for 30–60 minutes. After extraction, cool to room temperature and adjust the pH of the extract to 3.0–4.0 using an acid adjuster to promote the precipitation of some impurities. After settling and separation, the supernatant is filtered through a microfiltration membrane with a pore size of 100–500 nm to remove macromolecular colloids, suspended particles, and some protein impurities, yielding a clear microfiltration permeate.
Purification and Drying
The microfiltration permeate is transferred to a vacuum concentrator and heated to a suitable volume (1/5–1/10 of the original volume) to obtain a concentrated solution. The concentrated solution is then sent to a chromatography column for further impurity removal. A polyamide column is used to further remove pigments, tannins, and other polar impurities. The permeate is collected, and after passing inspection, it is sent to a spray drying tower for drying. Spray drying is performed with an inlet air temperature of 180–200℃ and an outlet air temperature of 80–100℃, resulting in an orange-yellow Rheum Palmatum Root Extract Emodin Powder.
