KINTAIBIO® betulin is primarily derived from the bark of birch and other plants in the Betulaceae family, with birch bark being the richest source, accounting for 20%–30% or more of the bark's dry weight. It is currently the most important natural raw material source for the industrial production of betulin. Birch is a deciduous tree, reaching up to 25 meters in height and 50 centimeters in diameter at breast height, with an oval crown. It is a light-loving and cold-resistant tree, widely distributed in the Greater Khingan Mountains, Lesser Khingan Mountains, Changbai Mountains in Northeast China, and high-altitude regions of Northern China.

How to make Betulin Powder?
Raw Material Screening and Pretreatment
KINTAIBIO® industrial production typically selects mature birch bark with a betulin content of approximately 15%–20%. After strict raw material acceptance, impurities and foreign matter are removed, followed by drying to maintain the moisture content within a suitable range. The bark is then pulverized to a uniform particle size using a pulverizing device to increase the contact area between the raw material and the extraction solvent, thereby improving subsequent extraction efficiency.
Ethanol Reflux Extraction and Concentration
The pretreated birch bark is added to an approximately 85% ethanol aqueous solution for reflux extraction. After reflux extraction, the extract is collected, filtered through multiple stages to remove plant residues, and then concentrated using a vacuum concentration device to recover ethanol and concentrate the extract. Once the concentrate reaches a suitable concentration, it is naturally cooled, and crude betulin gradually precipitates to form crystals. Solid-liquid separation yields crude betulin I, while ethanol is recovered and recycled, improving production efficiency and reducing energy consumption.
Alkali Treatment Purification and Crude Product Refining
The obtained crude betulin I was added to an 85%–95% ethanol aqueous solution at a weight ratio of 1:4–7. The solution was heated and stirred until completely dissolved, and then filtered while hot to remove insoluble impurities. Subsequently, a 3%–8% KOH or NaOH aqueous solution was added to the filtrate, with the volume controlled to be 0.9–1.1 times the total volume of crude product I and the ethanol solution. Alkali treatment further removed impurities such as resin, phenols, and pigments. The system was then cooled to 0–3°C to promote recrystallization of betulin. After filtration, crude betulin II with further improved purity was obtained.
Recrystallization Purification
Crude betulin II was added back to an ethanol aqueous solution with a mass concentration ≥90%, heated to fully dissolve, and then hot-filtered to remove trace amounts of insoluble impurities. The clear filtrate was collected and slowly cooled under strictly controlled cooling rates to allow betulin to crystallize uniformly, thereby further removing residual impurities and improving crystal purity and consistency. After crystallization, filtration and separation are performed to obtain high-purity betulin crystals.
Drying, Pulverizing, and Quality Inspection
After centrifugation to remove liquid, the high-purity betulin crystals are thoroughly dried using low-temperature vacuum drying equipment to remove residual solvents and moisture, ensuring product stability. The dried crystals are then pulverized, granulated, and sieved to obtain a white or off-white crystalline powder with uniform particle size and good flowability. Subsequently, comprehensive quality inspections are conducted on the product, including appearance, betulin content (HPLC), loss on drying, ash content, heavy metals, pesticide residues, microorganisms, and residual solvents, ensuring that the product meets the company's quality standards and customer requirements.
Packaging and Storage
