Optimization of Preparative HPLC for Separation and Purification of Asiaticoside and Madecassoside

Asiaticoside and madecassoside are key indicator compounds used for quality control of Centella asiatica and its related products. In this study, preparative high-performance liquid chromatography (HPLC) was employed to simultaneously isolate and purify these two compounds from Asiatic pennywort. Optimization of preparation parameters, including mobile phase composition, flow rate, injection volume, and detection wavelength, was carried out to achieve the best separation performance. A C18 column (50 mm × 200 mm, 5 μm) was used with a mobile phase consisting of methanol and water in a 60:40 (v/v) ratio. The flow rate was set to 100 mL/min, and the diode array detector (DAD) operated at a wavelength of 220 nm, with an injection volume of 1.5 mL. Under these conditions, asiaticoside and madecassoside were well separated from interfering substances within 20 minutes, and the product purity exceeded 98%. This method is rapid, efficient, and ensures high product purity, making it suitable for the preparation of asiaticoside and madecassoside reference standards.

Purification of Pennywort extract via Titration Method
