GTC-30(R) production process

(1) Production process Green tea is extracted with pure water, the extract is cooled, filtered, concentrated under reduced pressure, and spray-dried. This process is suitable for the production of GTC-30(R), and the yield is about 20%.

 

(2) Selection of extraction conditions Test extraction water consumption: 90-95 ° C, extraction time is 20 minutes, 12 times the water extraction rate is 67% (4E), 18 times the water extraction rate is 77%, that is, the increase in water volume increases the extraction rate . Extraction time: under the condition of 90-95°C, the extraction time of 18 times the amount of water is 30 minutes, and the extraction rate decreases (compared to 20 minutes), which may be that catechins are easily oxidized when heated for too long. Extraction times: 18 times the amount of water extracted twice, that is, 12 times of water (20min) and 6 times (15min), the second extraction rate can reach 29%, and the total extraction rate can reach 96%.

GTC-30(R) production process

 

Considering the unstable factors such as easy oxidation and mutarotation of polyphenols under high temperature conditions, 18 times the amount of water was used for extraction at 90-95°C for 20 minutes at a time. The longer the water extract stays at high temperature, the more likely it is that EGCG will mutate into GCG. The extraction rate of GCG is as high as 350% at 90-95°C for 20 minutes, and about 450% at 30 minutes. In addition, 30% ethanol extraction has the advantages of low temperature, less solvent consumption, and high extraction rate, and the GCG extraction rate is only 95%, which is only 25% to 30% of the same time at 90-95°C.

(2) Decaffeine test acid-base extraction test: get the green tea water extract, test according to the test method in Figure 2 respectively, concentrate under reduced pressure, vacuum dry under the same conditions, measure 4E (EC, ECG, EGC, EGCG) and c.f. (caffeine) content. Under acidic conditions (pH value 2-3), it is not only conducive to the extraction and transfer of catechins, but also to the separation of two phases. Moreover, extraction under acidic conditions can also reduce the transfer of caffeine to the organic phase and reduce the caffeine content in the product.

 

The comparison of pH adjustment experiments with citric acid, hydrochloric acid, acetic acid, and phosphoric acid shows that after adjusting the pH value to 2-3 with phosphoric acid, it is left for about 15 hours, and the extraction rate of catechin is high, the content of caffeine is low, and the yield is relatively high.
