Use dried roots or rhizomes of wild or cultivated Rheum rhaponticum Root as raw material. Cut the raw material into 2-5 mm slices using a cutting device, or crush it into 10-80 mesh powder using a pulverizing device to obtain the extract.

Ethanol Reflux Extraction

Collect the extract after each extraction, and combine to obtain a crude extract. Concentrate and dry the crude extract under reduced pressure at 30-50℃ and a vacuum of 0.01-0.1 MPa, or use spray drying to constant weight to obtain the total extract powder.

Preparation of the Two-Phase Solvent System

Mix methanol, water, n-butanol, and chloroform in a volume ratio of 1–2.7 : 1–4.3 : 0.5–1.5 : 1–3.8. Shake thoroughly and allow to stand at room temperature until the mixture naturally separates into layers. Collect the upper and lower phase solvents separately and sonicate each for 5–20 minutes to remove air bubbles. Use the upper phase solvent as the stationary phase and the lower phase solvent as the mobile phase in high-speed countercurrent chromatography (HSCLC).

Injection Solution Preparation

Dissolve 0.5–5 g of the total extract powder obtained in step two in 50–200 mL of mobile phase. Filter to obtain the injection solution.

HSCLC Separation

Pump the stationary phase into the separation tube of the HSCLC instrument while simultaneously rotating the separation tube forward at a speed of 350–550 rpm. When stationary phase overflows from the outlet of the separation tube, switch to pumping the mobile phase into the separation tube at a flow rate of 10–30 mL/min. When mobile phase overflows from the outlet of the separation tube, inject the injection solution into the injection coil, maintaining a constant mobile phase flow rate.

After the instrument runs forward for 70-100 minutes, collect the eluent A from this period and dry it to constant weight to obtain Rheum Rhaponticum Root Extract Desoxyrhaponticin. Continue running the instrument forward for 120-320 minutes, then switch the separation tube to reverse rotation at the same speed, maintaining a constant mobile phase flow rate.
