4.Physical and chemical identification
(1) The powder of this product is greenish yellow or yellow. The fibers are bright yellow, 16-38 μm in diameter, often in bundles, and the surrounding cells contain square crystals of calcium oxalate, forming crystal fibers; the cell walls containing crystals are lignified and thickened. Stone cells are bright yellow, sub-round or spindle-shaped, 35-128 μm in diameter, and some are branched, with sharp tips, thick walls, and obvious layered lines. The diameter of the square crystal of calcium oxalate is about 24 μm.
(2) Take 1g of this product powder, add 10ml of diethyl ether, shake, filter, evaporate the filtrate to dryness, add 1ml of glacial acetic acid to dissolve the residue, add 1 drop of sulfuric acid, let it stand, the solution becomes purple-brown.
(3) Take 0.1g of the powder of this product, add 5ml of methanol, heat to reflux for 15 minutes, filter, add methanol to the filtrate to 5ml, and use it as the test solution. Take another 0.1 g of Cortex Phellodendri as reference medicinal material, and prepare the reference medicinal material solution in the same way. Then take the berberine hydrochloride reference substance, add methanol to make a solution containing 0.5mg per 1ml, as the reference substance solution. According to the thin-layer chromatography (Appendix Ⅵ B) test, draw 1 μl of each of the above three solutions, and spot them on the same silica gel G thin-layer plate, and use benzene-ethyl acetate-isopropanol-methanol-concentrated ammonia test solution (6 :3:1.5:1.5:0.5) as the developing agent, put it in a developing cylinder saturated with ammonia vapor, develop it, take it out, dry it, and inspect it under an ultraviolet lamp (365nm). In the chromatogram of the test product, at the position corresponding to the chromatogram of the control medicinal material, a fluorescent spot of the same color appears; at the position corresponding to the chromatogram of the reference product, a yellow fluorescent spot of the same color appears.
5.Function
1). Antibacterial: Berberine is the active antibacterial ingredient of Phellodendron Phellodendri. In vitro tests are effective against Staphylococcus aureus, Pneumococcus, Bacillus diphtheria, Streptococcus viridans, and Shigella (except Sonnell's), etc. It is reported to be effective in Escherichia coli and Pseudomonas aeruginosa.

4). Anti-hepatitis: Phellodendron decoction 6.25% ~ 100% in vitro test, has inhibitory effect on hepatitis B antigen.
